T cell subsets in islets of young and aged mice. Islets (a,c) and spleens (b,d) were isolated from 3- and 18-month old mice. Islets from 10 mice pooled per sample, n = 1 sample/age group and spleens from 3 individual mice were assessed per age group. Cells were assessed for surface markers TCR αβ (TCR ab), TCR γδ (TCR gd), CD4, CD8 and NK1.1 after gating on FSC, SSC, viability (7AAD-), CD45+ and TCR+ population. a-b) Numbers in FACS plots represent the percent of cells in each selection as a function of the parent population. c-d Quantification as a percent of total TCR+ cells shown as mean ± SD for islet (c) and spleen (d).
