CTL heterozygous for SNX27 show enhanced expression of inflammatory and cytotoxic molecules. A Ex vivo-differentiated CTL from SNX27fl/fl or CD4-Cre-SNX27fl/+ mice were stimulated with soluble anti-mouse CD3 and CD28 (1 μg/ml) for the indicated times. Cell lysates were analyzed by Western blot using the depicted antibodies. GAPDH was used as a loading control. A representative blot and the quantification of p-ERK and p-rpS6 levels are shown (n = 3 mice). Values were normalized against GAPDH. Data are shown as mean ± SEM; two-way ANOVA with Bonferroni post-test was used for multiple comparisons; ns p > 0.05. B Ex vivo-differentiated CTL from SNX27fl/fl or CD4-Cre-SNX27fl/+ mice were stimulated with plate-bound anti-CD3 (2.5 μg/mL) and soluble anti-CD28 (1.25 μg /mL) for 24 h. Relative expression of the indicated genes was calculated by qPCR using de ΔΔCT method. β-actin was used as control. Data are shown as mean ± SEM; *p < 0.05. Two-way ANOVA with Bonferroni post-test was used for multiple comparisons. n = 9 mice. SNX Sorting nexin, CTL Cytotoxic lymphocytes, SEM Standard error of the mean
